I painted a frame over the weekend with Acrylic Enamel and ended up with a couple runs...can I wet sand them out and buff the shine back or will it need to be resprayed after sanding?

Yes it can be cut and buffed, if you used hardner in it.

Paint store sells a small square tool with a file type side on one side. We call it a "nib" here. It allows you to scuff off most of the drip before wet sanding, thereby preventing sanding off too much paint on each side of the drip, run, sag etc.

After sanding whats the best thing to use to bring the shine back?

Hit it with a little polishing compound and then some buff compound on a wool pad, just keep it SLOW and keep the heat down. We use the 3m stuff here, but I would recommend painting a test piece with the same paint, and testing the whole process on something less important just to be sure it's all going to work for you.
